    the most toxic members of the plant kingdom has had a varied and complex taxonomic history It has been described as the odd man out in the family because it is one of only a few umbellifers that produces alkaloids It also lacks well developed vittae in its mature fruits As such the genus has been variously treated in traditional taxonomic studies Moreover the relationships among its constituent species especially between the widespread weed C maculatum and its southern African congeners are obscure Molecular systematic studies to date have revealed that C maculatum comprises an isolated lineage within the apioid superclade a large and variable group of umbellifers containing tribes and other major clades of uncertain relationship To ascertain the phylogenetic placement of the genus within the apioid superclade and to assess infrageneric relationships we are analyzing nrDNA ITS and cpDNA sequences Preliminary results using ITS sequences indicate a sister group relationship between Conium and an expanded tribe Tordylieae an intriguing find since some of these plants share a similar phytochemistry and insect associates Further sampling of tribe Tordylieae and its allies is in progress The three species of Conium endemic to southern Africa C chaerophylloides C fontanum and C

    Abstract Detail Growth and Vegetative Development Burkart Graham 1 Harris Jeanne M 2 A Screen for ABA insensitive Mutants in Medicago truncatula The plant hormone Abscisic acid ABA mediates responses to abiotic stresses such as drought cold and high salinity Previous findings in our lab have shown abscisic acid to also play a role in nodule development as well as a novel role in lateral root growth in Medicago truncatula Contrary to findings in Arabidopsis ABA stimulates lateral root growth and formation in M truncatula rather than inhibiting it This suggests that ABA signaling in legume roots may be different than in non legume roots In order to better understand the function of ABA in nodulation and lateral root growth in M truncatula M2 seeds from an EMS mutagenized population were screened for insensitivity to ABA for germination We found a total of 61 putative ABA insensitive mutants representing 26 lines that germinated on media containing 50 uM ABA We are currently re screening the M3 generation Plants that re screen will have known and suspected ABA responsive genes sequenced and examined for mutations and the expression of these genes in the mutants will also be assessed In addition we

    in the Alaska cedar Callitropsis nootkatensis Layering a form of clonal growth where low lying branches root adventitiously may promote inbreeding and relatedness in long lived coniferous trees In this first microsatellite study of asexual reproduction in the Alaska cedar Callitropsis nootkatensis 485 trees from nine plots within three mountain ranges were surveyed for variation at five loci to identify clones compare levels of clonal growth and genetic diversity and estimate the contribution of clonal ramets to geitonogamy biparental inbreeding and relatedness We used a novel procedure that uses the ratio of three gene to two gene coefficients of identity to contrast the contributions of clonal versus non clonal individuals to population structure Clonal extent differed significantly among plots and was positively correlated with inbreeding while overall levels of genetic variation were invariable Clonal ramets contributed excessively to three gene to two gene coefficients of identity indicating that layering was responsible for local inbreeding and relatedness Despite adaptations that promote outcrossing including monoecy and tall stature the increased inbreeding due to Alaska cedar ramets concurs with the pervasive dogma that geitonogamy is a tolerated by product of other processes The genetic load accompanying inbreeding may contribute to the poor sexual

    to connect to database 13 20 37 SQL Statement is null or not a SELECT 13 20 37 Abstract Detail Intracellular Signaling Anderson Jeffrey C 1 Peck Scott C 2 A simple and rapid technique for stoichiometric detection of protein phosphorylation Recent advances in phosphoproteomic based approaches have rapidly increased the inventory of known phosphoproteins However most techniques for measuring the phosphorylation status of individual proteins remain technically demanding to perform e g two dimensional 2 D gel electrophoresis or require a priori knowledge of specific phosphorylation sites e g immunoblot detection with phospho specific antibodies Furthermore although phosphorylation can alter the migration of proteins in SDS PAGE gels this shift only occurs for a subset of phosphoproteins To overcome these limitations we developed a method for detecting phosphorylation that involves isoelectric focusing of proteins in a vertical mini gel format under denaturing conditions followed by immunoblot detection of the protein of interest The method is simple easily implemented with standard laboratory equipment and provides a rapid means to analyze the phosphorylation status of individual proteins In addition this approach provides a stoichiometric measure of phosphorylation making it possible to determine the extent to which a protein is phosphorylated Examples

    Environmental Physiology Patitucci Teresa 1 Feirer Russ 2 A Simplified Method for the Measurement of Proline in Stressed Tissues Higher plants produce the amino acid proline in the response to dehydration This compound provides intracellular protection against water loss a process of profound economic and agronomic importance In plants more primitive than gymnosperms little is known about the occurrence and role of proline during water stress or the genes or enzymes responsible for proline biosynthesis A focus of our work is the study of the occurrence of proline in a primitive plant s responses to water stress A commonly used spectrophotometric technique for the measurement of free proline in plant tissues Bates et al 1973 was modified to simplify the protocol and utilize smaller amounts of tissue Free proline levels were routinely determined in tissues weighing less than 100 mg fresh weight The utility of the technique was confirmed by the detection of elevated proline levels in osmotically stressed intact and in vitro grown Arabidopsis thaliana tissues Elevations of free proline have not detected in stressed fern tissues Intact and in vitro derived Ceratopteris richardii tissues were subjected to osmotic stress by a 24 hr exposure to 0 6 M

    design of most of these vectors limits them to the cloning of a single target gene in parallel to the expression of a selection gene from the same vector A vector cloning system was recently developed in which multiple expression cassettes cloned in small satellite plasmids are assembled into a binary plasmid using a set of rare cutting enzymes allowing the construction of up to seven expression cassettes in a single binary vector To overcome the problem of the very small number of commercially available rare cutting enzymes a set of zinc finger nucleases ZFNs and compatible plasmids were developed ZFNs are artificial restriction enzymes which can be custom designed to recognize and cleave specific 24 to 30 bp long sequences These enzymes are composed of a custom designed DNA binding domain fused to the cleavage domain of the Fok I endonuclease A set of different ZFNs were produced by de novo assembly of their DNA binding regions from overlapping oligos containing modified helices responsible for DNA triplet recognition and they were cloned into bacterial expression plasmids A compatible set of satellite plasmids was also developed in which each expression cassette was flanked by recognition sites of different ZFNs Following
